摘要
目的开发一种灵敏度高且简单快速的胶体金免疫层析检测分析系统,为人居环境中Der f1变应原的检测提供检测基础。方法采用柠檬酸三钠还原法制备胶体金颗粒。用胶体金标记的单克隆抗体建立免疫层析分析系统,用以检测Der f1。其检测效果与ELISA法相对照。结果建立的胶体金免疫层析检测方法灵敏度达50μg/L以上,特异性好,无交叉反应,与ELISA法相比较阳性符合率为94.1%,试剂稳定性可达6个月以上。结论胶体金法检测Der f1变应原特异性强、灵敏度高、简便快速,无需特殊仪器设备,具有广泛应用价值。
Objective To develop a sensitive, rapid and simple colloidal gold immunochromatography assay for detecting Der fl in living environment. Methods Colloidal gold was prepared with the tri-sodim citrate reduction method. Der fl was detected with immunochromatography. Rate of coincidence was compared with ELISA. Results Sensitivity of the established method was 50 μg/L,with good specificity,but without cross reaction;rate of coincidence was 94.1% compared with that of ELISA. Conclusion Gold immunochormatographic assay for Der fl is a method with a wide range of application,high sensitibity and good specificity, easy application, and that need no special equipment.
