紫杉醇对乳腺癌大鼠围手术期血儿茶酚胺及其他指标的影响

The Effects of Paclitaxel on the Plasma Level of Catecholamine and Some Immune Parameters during Perioperative Period in Rat Model of Breast Cancer

摘要 目的:探讨紫杉醇对乳腺癌大鼠模型围手术期血浆儿茶酚胺浓度以及部分免疫指标的影响。方法:选取SD大鼠70只,将walker-256肿瘤细胞接种于大鼠左侧腋下形成肿瘤结节,将存活并接种成功的50只大鼠随机分为4个实验组(分别为第1、2、3、4组)和1个对照组,每组10只。实验组静脉注射紫杉醇(25μg/g),对照组注射等量生理盐水,实验组分别于给药后第1、2、3和4周行肿瘤切除术。分别于术前及术后取静脉血测定肾上腺素和去甲肾上腺素水平、自然杀伤(NK)细胞计数、CD4+/CD8+细胞比值、肿瘤坏死因子(TNF)-α和白细胞介素(IL)-6水平。结果:各组术前和术后肾上腺素水平、NK细胞计数、TNF-α水平差异无统计学意义(P>0.05),第1、2组术前术后去甲肾上腺素浓度均高于对照组,CD4+/CD8+均低于对照组,第3组术前CD4+/CD8+低于对照组,第1~3组术前术后IL-6值均低于对照组,差异有统计学意义(P<0.01)。结论:静脉注射紫杉醇可造成乳腺癌大鼠以去甲肾上腺素水平升高为主的应激反应,同时对其免疫系统造成一定的抑制。 Objective:To investigate the effects of paclitaxel on the plasma level of catecholamine and some immune parameters during the perioperative period in rat model of breast cancer.Methods:Seventy SD rats were used to establish the model of breast cancer.Fifty rats,successfully formed tumor in the left armpit by planting walker-256 cells,were divided into 4 experimental groups and 1 control group randomly(n=10 for each group).Rats of experimental groups were received intravenous infusion of paclitaxel(25 μg/g),while rats in control group was given the same dose of normal saline.The tumor resection operation removal surgery was performed after 1,2,3 and 4w treatment in rats of experimental groups.The blood levels of epinephrine,norepinephrine,natural killer(NK) cell count,CD4+/CD8+ ratio,tumor necrosis factors-α(TNF-α) and interleukine-6(IL-6) were detected before and after operation in rats of groups.Results:There were no significant differences in epinephrine,TNF-α and NK cell counter before and after the operation in rats of five groups(P 0.05).But the level of norepinephrine was higher before and after operation in group 1 and group 2 than that of control group.The level of CD4+/CD8+ was lower before and after operation in group 1 and group 2 than that of control group.The ratio of CD4+/CD8+ was lower in group 3 than that in control group.The levels of IL-6 were significantly lower before and after operation in groups 1,2 and 3 than those of control group(P 0.01).Conclusion:Intravenous infusion of paclitaxel can induce significant stress response characterized by the increasing of plasma level of norepinephrine,and also can suppress the immune system.
