摘要
目的:探讨核转录因子(NF)-κBp65和基质金属蛋白酶(MMP)-9蛋白在非小细胞肺癌中的表达及其临床病理意义。方法:采用免疫组织化学方法检测95例非小细胞肺癌(NSCLC组)和27例癌旁正常肺组织(对照组)中NF-κBp65和MMP-9蛋白表达水平并分析其临床意义。结果(:1)NF-κBp65蛋白在NSCLC癌组织中阳性表达率44.2%(42/95),明显高于癌旁正常肺组织22.2%(6/27),差异有统计学意义(χ2=4.26,P<0.05)。(2)NF-κBp65在不同组织学分型、分化程度、TNM分期及有无淋巴结转移患者表达差异均有统计学意义(P<0.05或P<0.01),而不同年龄、性别及肿瘤大小患者表达率差异均无统计学意义(P>0.05)。(3)MMP-9蛋白在晚期(Ⅲ期)NSCLC组织中阳性表达率高于早中期(Ⅰ、Ⅱ期)者,差异有统计学意义(P<0.05);在腺鳞癌中阳性表达率显著高于腺癌和鳞癌(P<0.01),且随着肿瘤分化程度的降低,其阳性表达率逐渐升高,差异有统计学意义(P<0.05);不同年龄、性别及肿瘤大小患者MMP-9蛋白表达差异无统计学意义(P>0.05)。(4)NF-κBp65表达情况与MMP-9呈正相关(rs=0.217,P<0.05)。结论:NF-κBp65与MMP-9高表达可能与NSCLC的转移有关。
Objective：To investigate the clinical significance of protein expressions of nuclear factor kappa B（NF-κB） and MMP-9 in non-small cell lung cancer（NSCLC）.Methods：The expression levels of NF-κBp65 and MMP-9 protein were detected by immunohistochemical staining in NSCLC（n=95） and adjacent normal lung tissues（n=27）.Results：（1）The expression rate of NF-κBp65 protein was significantly higher in NSCLC（44.2%） than that of adjacent normal lung tissue（22.2%,χ2 =4.26,P 0.05）.（2）There were significant differences in expression levels of NF-κBp65 protein between the different tissue typing,clinical classification,clinical stage and metastasis（P 0.05 or P 0.01）,but there were no significant differences in expression levels of NF-κBp65 protein between the different age,gender and tumor size in NSCLC patients（P 0.05）.（3）The positive expression rate of MMP-9 protein was significant higher in patients of late-stage（Ⅲ） than that of early-and mid-stage（I-II）（P 0.05）.The expression rate of MMP-9 was higher in adenosquamous carcinoma than that of quamous cell carcinoma（P 0.01）,and the positive rate was gradually increased with the lower degree of tumor differentiation（P 0.05）.The expression rate of MMP-9 was not significantly correlated with the age,gender and tumor size in NSCLC patients（P 0.05）.（4） There was obviously positive correlation between NF-κBp65 and MMP-9（rs=0.217,P 0.05）,and both were correlated with the metastasis.Conclusion：The results indicate that NF-κBp65 and MMP-9 protein play a vital role in suppression of the metastasis of non-small cell lung cancer.
