复方甘草酸苷对HaCaT细胞表达AQP3的影响 被引量:7

摘要 目的观察复方甘草酸苷(compound glycyrrhizin,CG)对HaCaT细胞表达水通道蛋白3(aquaporin3,AQP3)的影响。方法不同浓度CG处理人角质形成细胞株HaCaT,MTT法检测CG对HaCaT细胞的增殖抑制作用,蛋白质印迹法(Western blotting method)分析CG处理后的HaCat细胞的AQP3表达。结果低浓度(31.25~62.5μL/mL)CG对HaCaT细胞生长没有明显影响,而125μL/mL CG则对HaCaT细胞的生长产生低毒,但AQP3的表达明显高于正常对照组。结论浓度(62.5~125)μL/mLCG可促进HaCaT细胞表达AQP3,而自125μL/mL开始出现细胞毒性,但此时AQP3表达至最高峰。 Objective To investigate the effect of compound glycyrrhizin(CG) on the aquaporin3(AQP3) expression in HaCaT cell cultured in vitro.Methods The keratinocyte cell line HaCaT was cultured with CG at various concentration,the cells proliferation were detected by MTT method.The expression of AQP3 was assayed by Western blotting method.Results CG at concentrations between 31.25 and 62.5μL/mL had no significant effect of the HaCaT cells,while the 125μL/mL of CG were generated on the low toxicity about the HaCaT cells,but the expression of AQP3 was significantly higher than the control group.Conclusion CG at concentrations between 62.5 and 125μL/mL can promote the expression of AQP3 in the HaCaT cells,and the 125μL/mL of CG began to appear cytotoxicity,but at this time to the peak expression of AQP3.
