黄芩苷对糖尿病肾病大鼠足细胞损伤的修复作用研究 被引量:19

摘要 目的:研究黄芩苷对糖尿病肾病(DN)大鼠血液AngⅡ及肾脏TGF-β1表达的影响,从细胞因子角度探讨黄芩苷通过修复足细胞损伤防治DN的作用机制。方法:应用链脲佐菌素(STZ)单次腹腔注射法建立DN模型,并随机分为DN模型对照组和黄芩苷治疗组,另设空白对照组。黄芩苷治疗组腹腔注射黄芩苷水溶液40 mg/kg,DN模型组每天腹腔注射注射用水1mL/只,干预42 d后,所有大鼠眼眶取血,用放射免疫法检测血浆AngⅡ;并称体质量、肾脏质量,计算肾脏指数;收集大鼠24 h尿液,测微量蛋白质、尿肌酐值;以3%戊二醛固定液固定右肾行电镜检查,4%多聚甲醛固定液固定左肾行病理学检测,观察肾小球足细胞形态改变及肾脏TGF-β1表达变化。结果:黄芩苷治疗组DN大鼠肾脏指数减小,与模型组比较,差异有统计学意义(P<0.05);尿微量蛋白含量明显减少,尿肌酐排泄量增加,与模型组比较,差异有统计学意义(P<0.05);电镜下足细胞损伤程度减轻;血浆AngⅡ降低,与模型组相比较,差异有统计学意义(P<0.05);TGF-β1表达数量减少,与模型组比较,差异有统计学意义(P<0.05)。结论:黄芩苷可通过降低血浆AngⅡ的含量来抑制TGF-β1的表达,改善肾小球足细胞损伤等非血压依赖机制减少蛋白尿,延缓DN进程。 Objective:To Study Effects of Baicalin Bai on AngII and TGF-β1 expression in diabetic nephropathy(DN)Rats,which to explore its preventive and therapeutic effect on treating DN by repaired of injured podocytesand from the cytokines.Methods:DN rat model was established substances in kidney of rat of streptozotocin.DN model rats were randomly divided into model group and Baicalin treatment group,Normal control group was set up additionally.Rats in Baicalin treatment group were injected Baicalin Bai solution 40mg/kg/day abdominally,rats in the model group were injected water 1ml/rat/day abdominally,Six weeks later,blood were drawn from eyes,AngII was measured by radioimmunoassay;the weight of their bodys and kidneys was monitored.Excretion amount of urine protein and creatinine in all rats were monitored for 24 hours.The right kidney tissues was fixed in 3% glutaraldehyde solution,and electronic scopy was performed.The left kidney tissues were fixed in 10% formalin solution,and pathological scopy was performed.Observe the morphology changes in podocytes and TGF-β1 expression in kidneys.Results:the kidney index decreased in the Baicalin treatment group,and there was significant difference compared to the model group(P0.05);Urine protein decreased and urine creatinine increased in the Baicalin Bai group,there was significant difference compared to the model group(P0.05);under electric scopy,incrassation injured podocytes;AngII in the blood decreased in the Baicalin treatment group,and there was significant difference compared to the model group(P0.05);ubule TGF-β1 expression in the Baicalin treatment group decreased significantly compared to the model group.Conclusion:Baicalin Bai could reduce AngII in the blood and inhibition TGF-β1 expression,to decrease Urine protein may be by non-blood pressure depend-dent mechanism,such as improveing injured podocytes,delay the progress of DN.
