MPT-NAG测定N-乙酰-β-D-氨基葡萄糖苷酶的应用价值 被引量:1

摘要 目的应用新型底物6-甲基-2-硫代吡啶-N-乙酰--βD-氨基葡萄糖苷(MPT-NAG)建立检测N-乙酰-β-D-氨基葡萄糖苷酶活力的紫外动力学新方法,用于尿液中NAG活性测定。方法基于NAG催化水解对硝基苯-N-乙酶-β-D-氨基葡萄糖苷生成有色的对硝基酚这一反应,对缓冲液离子强度、pH值、底物浓度等最佳反应条件及各种实验参数进行研究。用该法测定了多种病尿样中NAG活活性。结果该法最低检出限为1.5 U/L,线性范围可达250 U/L(r=0.998 9),批内CV为<3%,批间CV<5%,回收率为101.3%。结论本法灵敏度高,线性、精密度好,结果准确,且操作简便快速,适用于全自动生物化学分析仪操作。
