牛大肠杆菌油乳剂疫苗免疫的研究 被引量:4

摘要 应用K99、F41菌毛兼生性牛大肠杆菌分离株8511-2菌毛亚单位油佐剂灭活苗、8511-2全菌体灭活苗、 8511-2菌毛与菌体混合灭活苗给产犊前7周和4周的怀孕母牛免疫, 用玻片凝集试验检测母牛抗体水平;产犊后8-10h犊牛口服攻毒800亿菌/ml 8511-2株。结果表明:三种苗免疫母牛均能使犊牛获得一定保护,犊牛腹泻平均保护率分别为55.6%、89.1%、94.6%,对照组为16.3%;菌毛苗和混合苗效果优于全菌体苗。方差分析表明:不同菌苗之间以及不同免疫程序之间的K99免疫效果均存在显著差异,不同菌苗之间F41免疫效果差异显著;利用最小二乘法建立数学模型,结合动态趋势图分析母牛抗体效价的变化规律,表明:K99免疫效果优于F41,两次免疫效果优于单免。 Three oil adjuvant vaccines,8511-2 whole-cell bacterial vaccine, 8511-2 pili vaccine and compound vaccine, were produced with single E.coli which contained both F_41 and K_99 antigens isolated from diarrheic calves in 8511 farm. Pregnant cows were immunized with the three vaccines at seven and four weeks before they were delivered of calves. The antibody titters of the cows were detected. The calves at 8 to 10 hours after born were challenged, with 8×10~11 bacterium per ml by stain 8511-2 vaccine (K^+_99,F^+_41). The results showed that there was a clear protection in calves challenge experiments; the average protecting rates of calves were 55.6%, 89.1%, 94.6% and only 16.3% in control group. Pili vaccine and compound vaccine had an advantage over the whole-well bacterial vaccine. Statistical analysis and backward test demonstrated that the differentials of the immunity effection of K_99 was outstanding between the different vaccines and between the different immunal processes; the same result of F_41 was obstained between the different vaccines; K_99 was better than F_41 and immunity was superior to one immunity.
