摘要
草坪草的种类繁多,包含禾本科十几个属。本研究根据已报导的AsEXP1基因序列设计引物,扩增并测序了草坪草3个属共计6个品种植物材料约460 bp的AsEXP1基因片段。序列比对显示,不同草坪草品种间存在基因序列的差异,并呈现种属特征,即同属品种存在特定的碱基插入、缺失或替换等,同属品种间同源率超过97%,不同属品种间同源率在91%~94%之间。据此设计3对特异引物cAs3F/cAs3R、cAs4F/cAs4R和cAs5F/cAs5R,并利用6个主要草坪草属的32个草坪草品种进行了验证,结果表明,引物组合cAs3F/cAs3R可以鉴定剪股颖属品种,引物组合cAs4F/cAs4R能鉴定早熟禾属品种,而引物组合cAs5F/cAs5R只在羊茅属和黑麦草属中有扩增。本研究结果表明,通过AsEXP1基因片段序列多样性分析,可以较好的鉴别常见草坪草品种的种属亲缘关系,确定不同种属间的进化路线,为有效利用草坪草种质资源并进行分子育种提供理论依据。
Turf-grass species widely distributed in the world across more than ten genera in Poaceae.Based on the sequence of AsEXP1 gene,several pairs of primer were designed and used for surveying its homologous in 6 turf-grass varieties from 3 genera.The amplified DNA fragments about 460 bp were sequenced and compared.The alignment result showed the polymorphisms with special insertion,deletion,or substitution in each genus.The identity among species from same genus is over 97%,while within 91%~94% among the species from different genera.Three pairs of specific primers were designed for each genus,and made confirmation in 32 turf-grass species from 6 genera.Of them,primer-pair of cAs3F/cAs3R can be well used for distinguishing the bentgrass from the others,while cAs4F/cAs4R for bluegrass and cAs5F/cAs5R for fescue grass and ryegrass only.It revealed that the polymorphism of AsEXP1 gene can be well used for the determination of phylogenetic relationships and evolution pathway analysis in turf-grass.The results here display an effective tool in screening the grass germplasm for molecular breeding.
