摘要
目的:探讨增强型体外反搏(EECP) 对急性心肌缺血犬循环及局部组织中血管紧张素转换酶( ACE) 的影响。方法:结扎犬冠脉左前降支造成急性心肌缺血模型,用紫外分光光度法测循环及局部组织ACE 活性。结果:急性心肌缺血时循环ACE 持续上升,165 min 时,上升幅度趋于平缓,经反搏治疗后60 min ,未见对ACE 有影响,120 min 时ACE 下降,但仍高于正常水平。局部ACE 在缺血时均被激活,反搏120 min 后,各组织ACE 活性呈不同程度下降,左心室及肾仍高于正常水平,主动脉降至正常,肺部甚至降至正常以下。结论:急性心肌缺血可激活循环及局部ACE,体外反搏可能通过降低ACE 活性来参与对缺血心肌的保护作用。
AIM:To investigate the affect of enhanced external counterpulsation (EECP) on serum and local angiotensin converting enzyme(ACE) in dogs with acute myocardial ischemia(AMI). METHODS: 18 healthy hybrid dogs were randomly divided into control( n=6 ), ischemic( n=6 ) and EECP( n=6 ) group. Acute myocardial ischemia was induced by acute obstruct of left anterior descending coronary artery in ischemic and EECP group. The ACE activity in serum and tissue were measured by ultraviolet assay.RESULTS:The ACE activity in serum showed a markedly increase in ischemic group. But the increase of 105 min was much greater than that of 165 min. The ACE activity in serum was not significantly altered by treatment with EECP for 60 min compared with ischemic group. In 120 min, the ACE activity of EECP group decreased, but was still higher than normal. Local ACE was also activated by ischemia. EECP could suppress all local ACE in different degree. The ACE activity in aortic was suppressed to normal level. But in kidney and left ventricular, they were still higher than normal. The ACE activity in lung was even lower than normal. CONCLUSION:AMI increase local and circulating ACE. EECP can protect ischemic myocardium by suppressing both circulating and local ACE activity.
