摘要
核盘菌(Sclerotiniasclerotiorum)引起包括油菜在内的多种经济作物的菌核病。为筛选抗菌核病的作物品种及探讨防治该病的其它途径,获得足够的子囊孢子是极其重要的技术环节。本研究对大量获得子囊盘的方法进行了改进。首先从158个单菌核分离物中筛选出了6个能够在高温(20℃)下直接形成子囊盘的核盘菌菌株。其次对形成菌核的温度条件、形成菌核的营养基质及菌核形成期等因于对子囊盘产生的影响进行了研究。结果表明所筛选的菌株在灭菌胡萝卜片上培育1个月即可产生大量的成熟菌核,这些菌核在20℃下保湿10-30d即可形成大量的子囊盘和子囊孢子。菌株Zxd-1的菌核最容易前发。试验还表明按这一程序获得的子囊孢子长出的菌丝能感染油菜,抗(耐)性品种(中油821)和感病品种(Westar)对接种体的反应表现出明显的差异。
An improvement on mass production of apothecia of Sclerotinia sclerotiorum, a causal agent of sclerotinia stem rot of oilseed rape, was achieved by screening suitalbe strains and evaluating factors affecting carpogenic germination for this fungus. The results showed tha 6 out of 158 strains of S. sclerotiorum were screened out, sclerotia of which could germi-nate to form apothecia at 20℃ without any conditioning. The improved procedures for formation of apothecia included: (1) in-cubation of selected strains at 25℃ on autoclaved carrot slices for at least one month; (2)incubation of sclerotia on most sand at 20℃ f0r production of stipes; (3) transfer of sclerotia with stipes to an incubator of 20℃ with fluorescent light for develop-ment of apothecia. Strain Zxd-1 and other five strains of S. sclerotiorum were pathogenic to Brassica napus CVs Zhongyou 821 and Westar, and difference in tolerance to S. sclerotiorum between the two cultivare can be detected with the screened stains and Canadian cmmon clones as well.
