摘要
目的探讨干扰素（ＩＦＮｓ）对骨肉瘤细胞表达基质金属蛋白酶－２（ＭＭＰ－２）和组织蛋白酶－Ｄ（Ｃａｔｈ－Ｄ）的调控作用，并为临床上应用ＩＦＮｓ防治肿瘤转移提供一些实验基础。方法采用ＩＦＮ－α及ＩＦＮ－γ对体外培养的ＯＳ－７３２骨肉瘤细胞ＭＭＰ－２和Ｃａｔｈ－Ｄ表达进行诱导调节，并利用细胞酶联免疫吸附试验（Ｃ－ＥＬＩＳＡ）方法对其表达水平的变化进行半定量检测。结果ＩＦＮ。及ＩＦＮ－γ对ＭＭＰ－２和Ｃａｔｈ－Ｄ的表达均有显著的诱导降低作用（Ｐ＜０．０１），且诱导规律较为一致，即随ＩＦＮｓ浓度的增加其表达量随之逐渐下降。结论ＩＦＮ－α及ＩＦＮ－γ在一定程度上能抑制骨肉瘤的浸润转移潜能，在治疗骨肉瘤中有一定的积极作用。
Objective To explore the regulatory effects of IFNs on the expression of MMP - 2 andCath - D in osteosarcoma cell and provide experimental evidence for clinical treatment of osteosarcomametastasis. Methods The changes of MMP - 2 and Cath-D were half quantatively measured by Cell ELISA in a human osteosarcoma cell line OS - 732, treated with IFN - α and IFN -γ. Results The resultdemonstrated that both of IFNs were capable of significantly decreasing their expression (P < 0. 01 ) in OS- 732 in a dose dependent manner. Condusion The findings suggest that IFNs may suppress their metastaticpotential and play a role in treatment of the patients with osteosarcoma.
