摘要
目的:采用HPLC法测定盐酸安非拉酮血药浓度,旨在为盐酸安非拉酮在体内的血药浓度分析和法医学中毒检测提供方法。方法:色谱柱采用YWG- C18Φ4.6×150m m 10μ,流动相为甲醇- 0.01m ol/L,醋酸铵(35:65,用氨水调pH7.5),茶碱为内标,检测波长为254nm ,血浆样品应用SPE固相小柱萃取。结果:安非拉酮线性范围为(5~500)μg/L(r= 0.9994),最低检测浓度5μg/L,平均方法回收率为97.03% 。日内、日间RSD均小于5% 。结论:此方法稳定、可靠。
Objective: A HPLC method has been developed to determine the concentration of amfepramone hydrochloride in plasma. The study devoted to analysis of amfepramone hydrochloride in plasma and toxication in forensic practice. Methods: They were separated and determined on SPE column and Φ4.6×150mm 10μYWG C 18 column respectively, using methanol 0.1mol/L ammonium acetate (35∶65) as the mobile phase, theophylline(theocin) as internal standard, UV 254nm detector. Results: The ranges of catibration curves were 5~500μg/L( r =0.9994) and lowest detectable concentration of amfepramone hydrochloride was 5μg/L, while the average recovery of the methods was 97.03%. The RSD for inter day and intraday assays were all less than 5%. Conclusion: The method is stable and reliable. The way was suitable for the analysis of amfepramone hydrochloride in plasma and determination of this toxication in forensic practice.
