The Effect of IL 6 on the growth of prostate cancer cells in vitro and its modulation

摘要 目的 了解白细胞介素6(IL6) 对前列腺癌(prostate cancer,PCa) 细胞体外生长的影响及地塞米松(dexamethasone,Dex) 对此的调节。方法 采用ELISA 法测定PCa 细胞系PC3m 细胞IL6 的自分泌及Dex 处理后的分泌状况,并用结晶紫法观察IL6 、抗IL6 抗体和Dex 对PC3m 细胞生长的影响。结果 PC3m 细胞可自分泌IL6,IL6 无促进PC3m 细胞增殖作用( P> 0.05);抗IL6 抗体可抑制PC3m 细胞生长,细胞毒性由(14.84 ±1.25) % 递增至(41 .41 ±0 .80)% ,呈剂量依赖关系( P<0.05) 。Dex 不能减少IL6 的产生但可抑制PC3m 细胞增殖。细胞毒性由(14 .06 ±0.20)% 递增至(31 .25 ±1 .19)% ,呈浓度依赖关系( P< 0 .05)。结论 IL6 为PC3m 细胞的自分泌生长因子;Dex 可抑制PC3m 细胞增殖,但并非通过抑制IL6 的生成而发挥作用。 Objective\ To study the role of interleukin 6 (IL 6) in the pathogenesis of an androgen independent prostate cancer line (PC 3m) and the modulatory effect of dexamethasone. Methods\ The secretion of IL 6 by PC 3m cells treated without or with dexamethasone were measured by using a pretty sensitive and specific enzyme immunoassay (ELISA), and the effects of exogenous IL 6, anti IL 6 monoclonal antibody and dexamethasone on the proliferation of PC 3m cells were determined by applying crystal violet assay. Results\ PC 3m cells produced certain IL 6. Exogenous IL 6 had no obvious promotion on the proliferation of PC 3m cells ( P >0.05), while anti IL 6 monoclonal antibody could dose dependently inhibited the growth of PC 3m cells, with the cytotoxicity being increased from (14.84±1.25) % to (41.41±0.80) % ( P <0.05). Dexamethasone could not inhibit the production of IL 6, but dose dependently the proliferation of PC 3cm cells ( P <0.05) with the cytotoxicity being increased from (14.06±0.20) % to (31.25±1.19) %. Conclusion\ IL 6 acts as an autocrine growth factor for PC 3m, and dexamethasone can inhibit the growth of PC 3m cells by a mechanism independent of its effect on IL 6 mRNA expression.\;
