摘要
目的研究不同吸烟时间对大鼠气道上皮细胞4-羟基壬烯醛(4-hydroxynonenal,4-HNE)和转化生长因子β1(transforming growth factor-β1,TGF-β1)表达的影响,探讨吸烟所致慢性气道炎症性疾病中氧化应激和气道重塑的关系。方法雄性Wistar大鼠30只,随机分为对照组、短期吸烟组和长期吸烟组,每组10只。采用免疫组织化学法半定量检测各组支气管上皮细胞4-HNE和TGF-β1的表达。结果短期吸烟组(0.204±0.017)和长期吸烟组(0.264±0.022)4-HNE表达均较对照组(0.174±0.017)增高(P值均<0.05),长期吸烟组较短期吸烟组表达升高(P<0.05)。短期吸烟组(0.199±0.021)和长期吸烟组(0.247±0.025)TGF-β1表达均较对照组(0.175±0.018)增高(P值均<0.05),长期吸烟组较短期吸烟组表达升高(P<0.05)。TGF-β1和4-HNE表达水平呈正相关(r=0.935,P<0.01)。结论吸烟可以引起大鼠气道上皮细胞4-HNE和TGF-β1的表达水平升高,二者表达水平呈正相关,且随吸烟时间的增加表达水平增加。
Objective To explore the relationship between oxidative stress and airway remodeling of chronic airway inflammatory disease caused by smoking through researching effects of different continuous time of smoking on expression of 4-hydroxynonenal(4-HNE) and transforming growth factor-β1(TGF-β1) on airway epithelial cells of rats.Methods Thirty Wistar rats were randomly divided into a control group,a short-term smoking group and a long-term smoking group.The expression of 4-HNE and TGF-β1 were detected by Immunohistochemistry.Results The protein expression of 4-HNE in bronchial epithelialcells of the long-term smoking group(0.264±0.022) and the short-term smoking group(0.204±0.017) were higher than the control group(0.174±0.017)(all P〈0.05),with the long-term smoking group higher than the short-term smoking group(P〈0.05).The protein expression of TGF-β1 in bronchial epithelial cells of the long-term smoking group(0.247±0.025) and the short-term smoking group(0.199±0.021) were higher than the control group(0.175±0.018)(all P〈0.05),with the long-term smoking group higher than the short-term smoking group(P〈0.05).The expression protein of TGF-β1was positively correlated with the expression protein of 4-HNE(r=0.935,P〈0.01).Conclusions Smoking can result in the high protein expression of 4-HNE and TGF-β1 in time-dependent manner.
