摘要
目的探讨早期糖尿病视网膜病变(diabetic retinopathy,DR)大鼠的视网膜形态变化与血清血管内皮生长因子(vascular endothelial growth factor,VEGF)、内皮素(endothelin,ET)和一氧化氮(NO)的相关关系。方法 40只健康SD大鼠随机分为实验组和对照组,实验组腹腔注射链脲佐菌素建模成功后,按病程分为糖尿病2、4、6、8个月组;对照组腹腔注射等体积缓冲液,根据与实验组年龄匹配原则平均分为4组,每组5只。分别在第2、4、6、8个月检测血清VEGF、ET、NO含量,摘除眼球观察视网膜组织形态,应用SPSSl3.0进行统计学分析。结果实验组病程4个月始出现明显视网膜组织水肿,细胞排列紊乱,随病程延长逐渐加重。各实验组血清VEGF、ET含量较同鼠龄对照组高(P<0.05),糖尿病2个月组血清NO含量较同鼠龄对照组高(P<0.05),而糖尿病6个月组及糖尿病8个月组血清NO含量较同鼠龄对照组低(P<0.05)。随着病程延长,实验组血清VEGF、ET含量逐渐升高,NO含量逐渐降低(P<0.05)。实验组血清VEGF与ET含量成正相关(r=0.821,P<0.01),与NO含量成负相关(r=-0.814,P<0.01);血清ET含量与NO成负相关(r=-0.802,P<0.01)。结论 VEGF、ET和NO的含量与DR严重程度存在相关性,考虑血清VEGF、ET和NO含量可以间接反映DR大鼠视网膜病变的严重程度。
Objective To investigate the correlation between the retinal morphologic changes and the levels of vascular endothelial growth factor( VEGF), endothelin(ET) and nitric oxide(NO) in rats with diabetic retinopathy (DR). Methods Forty SD rats were randomly assigned into the experimental group and the control group. To establish the DR model, rats in experimental group were intraperitoneally injected with streptozotocin(60 mg/kg). Rats in control group were intraperitoneally injected with the same volume of buffer. Rats in both groups were randomly assigned into four subgroups with 5 rats in each subgroup. The animals were observed at 2, 4, 6 and 8 months respectively after the establishment of DR model. The serum levels of VEGF, ET and NO were measured,the eyeballs enucleated and the retinal morpha observed. The data were analyzed with SPSS 13.0 software. Results In the experimental group, retinal edema and cellular disorganization appeared at 4 months and became more severe when diabetes course extended. The serum levels of VEGF and ET in the experimental group were significantly higher than those in the control group ( P 〈 0. 05 ). The level of serum NO in 2-month experimental subgroup was significantly higher than that in the corresponding control subgroup(P 〈0.05 ). The level of serum NO in 6- and 8-month experimental subgroups was significantly lower than that in corresponding control subgroups (P 〈 0.05). With the progression of the disease, serum VEGF and ET levels increased gradually and the NO level decreased gradually (P 〈 0. 05 ). In the experimental group, the VEGF level was positively correlated with that of ET( r =0. 821 ,P 〈0.01 ) but negatively correlated with that of NO( r = -0.814 & r = -0. 802,P 〈0.01 ). Conclusion The levels of serum VEGF, ET and NO are correlated with the seriousness of DR and may predict the DR development indirectly.
