摘要
目的通过建立异常切应力诱导的兔颈总动脉粥样硬化(As)模型,观察血管内皮生长因子受体2(VEGFR-2)在颈总动脉血管壁的表达分布。方法 26只新西兰兔行右颈总动脉硅胶管套环后,随机分为对照组和高胆固醇组。全自动生物化学分析仪测定兔血浆中甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDLC)和低密度脂蛋白胆固醇(LDLC)含量;HE染色观察兔颈总动脉病理组织学改变;免疫组织化学法检测兔颈总动脉血管壁VEGFR-2的表达分布。结果 9周末,高胆固醇组TC、HDLC、LDLC水平高于对照组(P<0.05);颈总动脉HE染色显示高胆固醇组套环远心段和近心段有斑块形成,且近心段斑块更加明显,并可见大量泡沫细胞;免疫组织化学显示对照组颈总动脉血管壁内膜有少量VEGFR-2蛋白表达,高胆固醇组套环远心段和近心段内膜VEGFR-2的蛋白表达增多,其中近心段和远心段斑块内也有少量VEGFR-2蛋白表达。结论 VEG-FR-2在异常切应力诱导的兔颈总动脉粥样硬化病变中有表达,其主要分布在套环部位的远心段和近心段。
Objective To investigate the expression of VEGFR-2 in common carotid artery atherosclerosis induced by abnormal shear stress in New Zealand rabbits.Methods After assembled silastic collars around the right common carotid arteries of all the New Zealand White rabbits,they were randomly divided into two groups,control group(n=13) and high-cholesterol group(n=13).The control group was given a normal control diet and the high-fat group was fed a high-cholesterol diet(2% cholesterol wt/wt).All rabbits were euthanized at the end of the 9-week experiment.Plasma triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDLC) and low density lipoprotein cholesterol(LDLC) were determined by commercially enzymatic methods.The pathological changes of rabbit arteria carotis was examined by HE staining.VEGFR-2 protein distribution and expression was analyzed by immunohistochemistry.Results Plasma TC,HDLC and LDLC concentrations were markedly increased in high-cholesterol group.Plaques were identified to exist in the upstream from the cast obviously more than downstream from the cast in staining intensity in high-cholesterol group with HE staining.From immunohistochemistry,the expression of VEGFR-2 in the control group was only in the intima of blood vessel and the quantity was very little,but in the other group there was increasing in the expression of VEGFR-2 in the upstream and downstream from the cast of arteria carotis.Conclusion VEGFR-2 protein was expressed in common carotid artery atherosclerosis induced by abnormal shear stress in New Zealand rabbits and located in the upstream and downstream from the cast of arteria carotis.
