摘要
目的探讨微管相关蛋白轻链3(LC3)及自噬基因Beclin-1在低氧性肺动脉高压大鼠肺组织中的表达及其意义。方法成年雄性SD大鼠30只,随机均分为低氧组和常氧组。建立低氧性肺动脉高压模型,采用蛋白质免疫印记法分别检测两组大鼠肺组织中LC3含量及Beclin-1蛋白的表达。结果低氧组肺组织中LC3及Beclin-1蛋白含量较对照组显著增高(P<0.05)。结论低氧性肺动脉高压大鼠的自噬途径被激活,可能参与了低氧性肺动脉高压的发病过程。
Objective To explore the expressions of autophagy-related gene Beclin-1 and microtubule-associated protein light chain 3(LC3) in rats with hypoxic pulmonary hypertension.Methods Thirty male SD rats were randomly divided into 2 groups of control group and hypoxic group.Rats in hypoxic group was placed into a hypoxia [(10.0±0.5)%] oxygen chamber.Right heart catheterization was placed to measure pulmonary arterial pressure.The expressions of Beclin-1 and LC3 of lung tissues were detected by Western blot.Results The expressions of Beclin-1 and LC3in the lungs were higher in hypoxic group than those in control group(P〈0.05).ConclusionAutophagy pathway is activated in rats with hypoxic pulmonary hypertension,which may play an important role in the occurrence of hypoxic pulmonary hypertension.
