摘要
目的对分离自硇洲岛黄海葵(Anthopleura xanthogrammica)样品的126株细菌进行抗菌活性筛选和初步分类鉴定。方法以8种微生物(3株革兰阳性菌、3株革兰阴性菌和2株真菌)作为指示菌,采用琼脂扩散法筛选抗菌活性菌株,采用基于16S rRNA基因序列的系统发育分析和生物学特性研究对其中抗菌谱较广且抗菌活性稳定的菌株进行初步分类鉴定。结果 67株菌具有抗菌活性,阳性率为53.2%,抗菌谱较广且活性较强的9株,占受试菌株总数的7.1%。经过形态观察、生理生化特征及基于16S rRNA基因序列的系统发育分析表明,9株具有较强抗菌活性的菌株分别属于芽孢杆菌属(Bacillus)、盐芽孢杆菌属(Halobacillus)、短杆菌属(Brachybacterium)、拟诺卡菌属(Nocardiopsis)、盐单胞菌属(Halomonas)、假交替单胞菌属(Pseudoalteromonas)和弧菌属(Vibrio)菌株。结论分离自中国南海的黄海葵相关微生物抗菌活性菌株具有丰富的类群多样性。
Objective To investigate the Screening and preliminary identification of 126 antimicrobial strains isolated from Anthopleura xanthogrammica collected from the area of the Naozhou island in the South China Sea. Methods The antimicrobial activity of the strains were tested using the agar diffusion method employing eight indicator strains including Sarcina lutea, Staphylococcus aureus, Escherichia coli, Proteus vulgaris, Bacillus subtilis, Candida albican, Asperillus niger and Enterobacter aerogenes. The preliminary identification of the strains were investigated by using phylogenetic analysis based on 16S rRNA gene sequence comparisons. Results The results showed that 67 strains out of the 126 strains exhibited antibacterial activity, in which 9 strains exhibited strong antibacterial activity. Based on the results of morphologic characteristics, physiological and biochemical properties and phylogenetic analysis of 16S rRNA gene, 9 strains belonged to the genera Bacillus, Halobacillus, Brachybacterium, Nocardiopsis, Halomonas, Pseudoalteromonas and Vt'brio respectively. Conclusion The results presented here showed that there is abundant diversity of population of the antimicrobial stains isolated from Anthopleura xanthogrammica collected from the South China Sea.
