摘要
目的研究高温、高强度和阳光中紫外线照射下对士兵泪液中基质金属蛋白酶-3(MMP-3)及组织金属蛋白酶抑制剂-1(TIMP-1)表达的影响。方法选择训练士兵共30人,分别于训练前、后收集泪液和抽取外周血。采用酶联免疫吸附测定法检测训练前、后士兵泪液中MMP-3和TIMP-1蛋白水平的表达;以及检测外周血清中促肾上腺皮质激素(ACTH)、去甲肾上腺素(NE)、活性氧(ROS)-氧化氮(NO)、丙二醇(MDA)和超氧化物歧化酶(SOD)的表达水平;采用非接触眼压计对士兵进行训练前、后的眼压测量。结果士兵训练前、后的眼压比较无统计学意义(P〉0.05);士兵训练前、后泪液中MMP-3和TIMP-1表达水平分别为(0.068±0.016)、(1.105±0.169)、(0.055±0.006)、(1.277±0.102)ng,两组训练前、后比较均有统计学意义(P〈0.05);士兵训练前、后血清中ACTH,NE,ROS,NO,SOD表达水平分另0为0.223±0.088,0.100±0.032,0.231±0.072,0.106±0.012,0.111±0.015;0.393±0.099,0.172±0.057.O.331±0.117,0.116±0.017,0.121±0.013,训练前、后比较均有统计学意义(P〈0.05),士兵训练前、后血清中MDA表达水平比较无统计学意义(P〉0.05)。结论高温、高强度和阳光中紫外线辐射下训练会对机体产生明显影响,机体产生自由基增多,使机体处于慢性氧化应激状态,泪液中MMP-3表达下降,TIMP-1表达升高。
Objective To investigate the changes and the role of matrix metalloproteinase (MMP)-3 and tissue inhibitor ofmetalloproteinase (TIMP)-1 in soldier tears drill under high temperature, intensive exercitation and UV radiation of sunlight. Methods The numbers of subject were 30 drilled soldiers. Tears and blood from peripheral vein were obtained and tested before and after drill. Fifteen microliters of tears was collected by microcapillary tubes from each eye. MMP-3, TIMP-1, adrenocorticotropic hormone (ACTH), nore- pinephrine (NE), reactive oxygen species (ROS), nitric oxide (NO), malondialdehyde (MDA) and superoxide dismutase (SOD) were detected by Enzyme Linked Immunosorbent Assay (ELISA). Intraocular pressure (IOP) was measured by non-contact tonometer (NCT) before and after drill. Results No statistical differences in IOP were noted between before and after drill (P 〉0.05); MMP-3 and TIMP-1 in tears was 0.0683±0.016, 1.105 ±0.169; before a drill and 0.055 3± 0.006, 1.277 ±0.102 after a drill, the differences of MMP-3 and TIMP-1 in tears between before and after drill were statistical significantly (P 〈0.05); The serum level of ACTH, NE, ROS, NO and SOD was 0.223± 0.088, 0.1003± 0.032, 0.231± 0.072, 0.1063±0.012, and 0.1113± 0.015 respectively before a drill and 0.3933± 0.099, 0.172± 0.057, 0.331± 0.117, 0.1163± 0.017, and 0.1213± 0.013 respectively after a drill, the serum level ofACTH, NE, ROS, NO and SOD after a drill were significantly higher than those before a drill (P 〈0.05); No significant difference were detected in the serum level of MDA between before and after drill (P 〉0.05). Conclusions Organism could be affected, and produce more flee radical and turn to stress state due to drill under high temperature, intensive exercitation and UV radiation of sunlight. Oxidative stress leads to MMP-3 reduce and TIMP-1 rose in tears.
