泰和乌骨鸡肉模拟胃肠道消化液中ACE抑制肽的分离纯化及结构鉴定 被引量:6

摘要 研究泰和乌骨鸡肉在胃肠道消化过程中的降血压活性,并鉴定出其中确定具有降血压活性的肽序列。采用体外模拟胃肠道消化法,测定了不同阶段消化液对血管紧张素I转化酶(ACE)的抑制作用,并选择其中的小肠道消化液,对其进行超滤、制备型反相高效液相色谱、凝胶柱色谱分离纯化,最后通过HPLC/Q-TOF MS鉴定出一种新型ACE抑制肽G lu-Pro-Leu-Tyr-Tyr。 The present study aimed to investigate antihypertensive activity of the digested substances of Taihe Black-bone silky fowl(Gallus gallus domesticus Brisson) by simulated gastrointestinal digestion system,and also to identify the key bioactive peptide with antihypertensive activity.A simulated gastrointestinal digestion in vitro mode was used.The digest of different stage was characterized and angiotensin I converting enzyme(ACE)-inhibitory activity was determined.The final 360-minute digest was selected for separation and purification.After ultrafiltration,preparative RP-HPLC chromatography and gel filtration chromatography,a novel ACE-inhibitory peptide Glu-Pro-Leu-Tyr-Tyr was identified by HPLC/Q-TOF MS.
