摘要
采用反相高效液相色谱法建立一种同时测定金边瑞香提取物中活性成分紫丁香苷、瑞香素和伞形花内酯含量的方法。以乙腈-甲醇-0.1%磷酸水溶液为流动相,流速为1.0 mL.min-1,色谱柱C18,检测波长为225 nm,柱温20℃。结果表明:紫丁香苷(syringin)、瑞香素(daphnetin)和伞形花内酯(umbelliferone)浓度与峰面积呈良好的线性关系,线性范围分别为0.0252-0.126μg,r=0.999 3;0.263 4~1.317μg,r=0.999 9;0.203 4~1.017μg,r=0.999 9。加样平均回收率(n=3)分别为107.04%,RSD=0.44%;105.59%,RSD=0.96%和98.18%,RSD=0.17%。该方法简便,准确,重现性好,适用于金边瑞香中紫丁香苷、瑞香素和伞形花内酯的含量测定。
A HPLC method was approached for the simultaneous determination of syringin,daphnetin and umbelliferone in Daphne odora Var.Margirmt extract.Chromatographic analysis was carried out by using acetonitrile-methanol-0.1% phosphoric acid as the mobile phase on a C18 column with gradient elution at a flowed of 1 mL·min-1 and detection wavelength of 225 nm under 20 ℃.A good linear relationship between concentrations and peak area of syringin,daphnetin and umbelliferone was absered.The linear ranges were 0.025 2~0.126 μg,0.999 3;0.263 4~1.317 μg,0.999 9 and 0.203 4~1.017 μg,0.999 9,respectively.The average recoveries were 107.04%,105.59% and 98.18%(n=3),respectively.The method was simple,accurate,good repetition rate and reliable for quantification of syringin,daphnetin and umbelliferone in Daphne odora Var.Margirmt extract within 11 min.
