摘要 目的探讨湖北恩施地区汉族人群TCF7L2基因多态性与2型糖尿病及发病风险的关系。方法采用PCR-RFLP法对湖北恩施地区258名新诊断2型糖尿病患者及239名健康对照者进行基因多态性检测,并进一步分析2型糖尿病及发病危险和TCF7L2基因多态性的关系。结果①2型糖尿病组的腰围、体重指数、空腹血糖、HbA1 c和甘油三酯明显高于正常对照组。②2型糖尿病组和对照组相比TCF7L2的TT基因型分布频率显著升高,且在人群中,携带TT基因型的患病风险是CC基因型的2.14倍。③Logistic多元回归分析显示,HbA1 c、BM I、TCF7L2基因多态性和TG与2型糖尿病的发生相关明显。进一步对2型糖尿病不同基因型的HbA1 c、BM I和TG水平进行分析,发现在3个基因型之间这些因素存在差异。结论在湖北恩施地区汉族人群中,TCF7L2基因多态性和2型糖尿病发生风险相关。 Objective To investigate the relationship between TCF7L2 polymorphism with type 2 diabetes and related factors. Methods The TCF7L2 polymorphism of 258 newly diagnosed type 2 diabetic patients and 239 healthy controls were detected by PCR-RFLP assay,and the relationship between TCF7L2 polymorphism with type 2 diabetes and its related factors was analyzed. Results Compared with the control group, the levels of waist circumference, BMI, FBG, HbAlc, TG and the frequency of Tr genotype were significantly in- creased in T2DM group, and the risk of type 2 diabetes in TT genotypo was 2.14 times as high as that in CC genotype. Logistic multifactor analysis found that there was notable correlation between type 2 diabetes incidence with HhAlc, BMI ,TCF7L2 polymorphism and TG, further analysis showed the significant differences of HhAlc, BMI and TG among different genotypes. Conclusion TCF7L2 polymorphism is associated with the incidence of type 2 diabetes among the Han population in enshi.
