生长激素释放肽对心衰大鼠心肌的影响

The influence of GHRP to the myocardium on the heart failure rats

摘要 目的研究生长激素释放肽(GHRP)对心衰大鼠心肌的影响,探讨GHRP在心衰(CHF)中的作用,为临床更好地治疗CHF提供理论依据。方法选取健康Wistar雄性大鼠30只随机分为三组,每组10只。采用腹腔注射阿霉素(ADR)方法建立大鼠CHF模型,并采用:1)监测血流动力学变化;2)采用HE染色法观察心肌细胞形态变化;3)电镜下观察心肌细胞形态变化。结果 1)模型组与正常组比较血流动力学和心肌组织发生了显著变化,而治疗组干预后的指标接近正常组;2)正常组线粒体结构正常,闰盘结构完好;模型组少数区域有线粒体空泡样变,闰盘结构正常;治疗组许多区域有线粒体肿胀、空泡样变,闰盘电子密度降低。结论 GHRP具有保护和改善心衰大鼠心功能的作用。 Objective To study the effect of GHRP to the myocardium on the heart failure rats, and explore the effect of GHRP on CHF in order to offer theoretical arguments for clinic to cure CHF much better. Methods 30 healthy Wistar female rats were selected and divide randomly into three groups, and 10 in each group. We set up the CHF models by injecting ADR and we used: (1)Useful methods such as monitoring the hemodynamic change; (2) HE dyeing of myoeardium of rats to observed morphological changes of myocardial cells; (3)Electron microscope to observed morphological changes of myocardial cells. Results (1)Compared with normal group, the changes of hemodynamics and myocardial of model group were significant; But treatment group had no obvious change; (2)normal group kept normal mitochondrial structure, and intact of intercalated disc structure; model group had mitochondrial vacuolar degeneration in a few regions, intercalated disc structure had no change; in many areas of treat- ment group mitochondria swelling, and vacuolar degeneration, and intercalated disc lower electron density in many regions. Conclusions GHRP may protect and improve the heart function of CHF rat.
