乳糖诱导重组大肠杆菌表达胆固醇氧化酶的研究 被引量:2

Expression Cholesterol Oxidase Gene in Recombinant Escherichia coli Using Lactose as Inducer

摘要 采用乳糖诱导胆固醇氧化酶(COD)基因在大肠杆菌BL21(DE3)中表达,研究了培养基成分、乳糖浓度、诱导时间和诱导温度对胆固醇氧化酶表达的影响。结果显示,在对诱导条件进行优化控制的前提下,胆固醇氧化酶酶活达到15.2 U/mL。研究结果为乳糖作为诱导剂最终应用于重组基因工程药物的工业化生产提供了有益的参考和借鉴。 Lactose was used to induce cholesterol oxidase(COD) expression in Escherichia coli BL21(DE3) in this manuscript.Effect of different nutrient and environmental conditions,including media ingredients,lactose concentration,lactose addition time,and the culture temperature on COD expression were carefully investigated.Under the optimum induced conditions,the maximal activity of COD reached at 15.2 U/mL.The results confirmed that lactose could be used as an inducer in the fermentation process.
