摘要
目的 探讨单胺氧化酶 B( M A O B) 抑制剂对衰老模型小鼠的抗氧化作用。方法 分别采用化学发光法、二硫对硝基苯甲酸( D T N B) 法、过氧化氢酶试剂盒法和荧光法检测丙炔苯丙胺(selegiline) 和2己基 N甲基丙炔胺(2 Hexyl Nmethylpropargylamine ,2 Hx M P) 对 D半乳糖诱导的衰老模型小鼠(11 只) 脑和肝铜/ 锌超氧化物歧化酶( Cu/ Zn S O D) 、谷胱甘肽过氧化物酶( G S H Px) 、过氧化氢酶活力和脂褐素含量的影响。结果 与衰老模型组相比较,selegiline(025 mg/kg) 组(12 只) 和2 Hx M P(10 mg/kg) 组小鼠(12 只) 脑、肝 Cu/ Zn S O D 和肝过氧化氢酶活力均高于衰老模型组( P< 001) ,肝脂褐素水平低于衰老模型组( P< 005) ,2 Hx M P 组脑( P< 001) 和肝( P< 005) G S H Px 活力也高于衰老模型组;2 Hx M P 组脑、肝 G S H Px 和肝 Cu/ Zn S O D 活力均高于selegiline 组( P< 005) 。结论 M A O B 抑制剂可提高衰老模型?
Objective The purpose of this study was to investigate the antioxidative mechanisms of monoamine oxidase B (MAO B) inhibitors in aging model mice induced by D galactose Methods The experimental mice were injected with D galactose 80 mg/Kg to produce a model of aging The effects of selegiline (0 25 mg·Kg -1 ) and 2 Hexyl N methyl propargylamine (2HxMP,10 mg·Kg -1 ) on the activities of Cu/Zn superoxide dismutase (Cu/ZnSOD), glutathine peroxidase (GSH Px), catalase and lipofuscin content in the brain and the liver were measured using chemiluminescence method, dithio bis 2 nitrobenzonic acid (DTNB) method, catalase detection kid and fluorescentmetric method, respectively Results In comparison with aging model group, the higher activities of Cu/ZnSOD in brain and liver and catalase in liver, but the lower content of lipofuscin in liver were observed both in 2HxMP group and in selegiline group; and the activities of GSH Px in brain and liver were also higher in 2HxMP group In addition, the Cu/ZnSOD level in liver and GSH Px in brain and liver were higher in 2HxMP group than in selegiline group Conclusions It is suggested that MAO B inhibitors could increase the activity of antioxidase and decrease the lipid peroxide level, which might play an important role in the prolongation of life span
