摘要
目的:观察乙醇对于Na^+依赖高亲和摄取L-[~3H]谷氨酸(预孵育4main+孵育2min)的影响。方法:采用4个鼠龄段大鼠大脑皮层突触体。结果:大剂量(包括522mmol·L^(-1)及348mmol·L^(-1)乙醇对4个鼠龄段大鼠大脑皮层突触体摄取谷氨酸均呈显著抑制效应;中等剂量(174mmol·L^(-1))乙醇在新生、成年及老年大鼠也见一定抑制性效应,而对于青少年大鼠没有显著影响;低剂量(87mmol·L^(-1)以下)乙醇对于4个鼠龄段大鼠均没有明显效应。结论:乙醇特别在高浓度下对大鼠大脑皮层突触体高亲和摄取谷氨酸存在明显抑制效应,这会使细胞外谷氨酸水平增高,因而有可能在急性酒精中毒所致神经精神障碍的发生中起作用。
Aim: To study acute effect of ethanol on the Na+-dependent high affinity uptake of excitatory neurotransmitter L-glutamate acid. Methods: The assay of 3[H]-L-glutamate uptake was conducted on the cerebral cortex synaptosomes of rats at different ages. Results: Ethanol at high concentrations (both 348 mmol·L-1 and 522 mmol·L-1) was found inhibiting L-glutamate uptake in synaptosomes of newborn, male adolescent, male adult and male old rats after 4 min preincubation and 2 min incubation. Ethanol in the middle level (174 mmol·L-1) also decreased L-glutamate uptake in synaptosomes of newborn, male adult and male old rats, but had no effect on those of male adolescent rats. Low doses of ethanol (either 87 mmol·L-1 or 17. 4 mmol·L-1)were found having no effect on L-glutamate uptake in synaptosomes of all rats at different ages. Conclusion: Ethanol, especially at high concentration, might inhibit the high affinity L-glutamate uptake into nerve terminals, inducing high level of extracellular L-gluramate, that might be involved in the development of acute ethanol intoxication.
