摘要
目的观察维生素C对链脲佐菌素(STZ)诱导糖尿病大鼠肾缺血-再灌注损伤的影响。方法 32只SD大鼠随机均分为四组:A、B、C组为糖尿病大鼠,B、C组行缺血-再灌注处理,A、B组每天腹腔注射生理盐水0.5 ml,C组注射维生素C 200 mg/kg(0.5 ml),D组为空白对照组。观察血糖、尿素氮(BUN)、肌酐(Cr)、丙二醛(MDA)、超氧化物歧化酶(SOD)、细胞色素C(Cytc)、半胱天冬酶-3(caspase-3)的变化。结果 B组BUN、Cr、MDA含量较C组明显升高(P<0.05),SOD的活性明显降低(P<0.05)。C组caspase-3和Cytc的表达明显减少,肾小管损伤、坏死显著减轻。结论维生素C能减轻STZ诱导糖尿病大鼠肾缺血-再灌注损伤的氧化应激,抑制细胞的凋亡及改善肾功能。
Objective To investigate the protective effects of ascorbic acid on renal ischemia/ reperfusion in STZqnduced diabetic rat. Methods Thirty-two SD rats were randomly divided into four groups with 8 rats each. The rats in group A, B, and C were induced diabetes by STZ,group B anct group c were exposed to isehemia-reperfusion. The rats in group A and group B were given norma sailne 0:5 ml/d i. P. for 15 days, group C were given ascorbie add 200 mg.kg-1 .d-3i.P. for 15 dfiys, 'gf0Up D waS control. Results Compared with those in group B, the concentration of serum BUN,SCr and MDA were significant higher in group C(P〈0. 05). The SOD level in group B was 0Diious I6wer than that in group C(P,〈O. 05): In group C the tubular cell damage was lesser, the expressions of Cytc and caspase-3 were lower in group (3, Conclusion Vc treatment Can reduce renal injury and improve renal function by reducing lipid oxidation and cell apoptosis caused by IR in STZinduced diabetic rats.
