RASAL1基因在胃癌组织中的表达及临床病理意义 被引量:1

Expression of RASAL1 Gene in Gastric Cancer and Its Clinicopathological Significance

摘要 [目的]研究RASAL1基因及蛋白在胃癌组织中的表达,并分析其与胃癌临床病理特征的关系,初步探讨RASAL1在胃癌发生发展中的意义。[方法]收集50例胃癌及正常胃组织标本,采用RT-PCR方法检测组织中RASAL1mRNA表达,WesternBlot及免疫组织化学方法检测RASAL1蛋白的表达;并结合胃癌的临床病理资料作相关统计学分析。[结果]①RT-PCR检测显示胃癌组织中RASAL1mRNA表达的平均吸光度值为0.51±0.15,而正常胃组织的平均吸光度值为0.64±0.12,胃癌组织的RASAL1mRNA显著低于正常胃组织(P=0.0004)。②WesternBlot分析表明,胃癌组织RASAL1蛋白表达的平均吸光度值为0.52±0.22,而正常胃组织的平均吸光度值为0.68±0.23,前者显著低于后者(P=0.003)。③免疫组化结果表明,RASAL1蛋白主要在胞浆中表达;50例胃癌组织中RASAL1蛋白的表达分别为12例(-)、23例(+)、13例(++)、2例(+++);而在50例正常胃组织中分别为16例(++)、34例(+++);胃癌组织中RASAL1表达较正常胃组织降低(P<0.05)。④RASAL1蛋白的表达与胃癌患者性别、年龄无显著相关,而与肿瘤大小、分化程度、侵袭深度、淋巴转移及TNM分期相关,随病情进展呈下降趋势。[结论]RASAL1基因在胃癌组织中表达明显下调,且与分化程度及进展阶段相关,提示其可能参与了胃癌的发生发展过程,是胃癌的抑癌基因之一。
