金属蛋白酶多抗原表位DNA疫苗诱导免疫应答的研究 被引量:3

摘要 目的研究信号肽在真核表达系统中对金属蛋白酶多抗原表位DNA疫苗诱导小鼠免疫应答影响。方法人工合成含有金属蛋白酶多个抗原表位的DNA序列,将连接信号肽与不连接信号肽的该序列分别连接pIRESneo表达载体,构建的2种质粒免疫小鼠,ELISA法、出血中和法检测免疫后效价。结果 2种质粒诱导小鼠产生的抗体效价10-2为阳性,均能有效减轻金属蛋白酶引起的出血。结论信号肽的有无对金属蛋白酶DNA疫苗在小鼠体内表达没有影响。 We aimed to research the influence of signal peptide on the immune responses induced by metalloproteinases multi-antigenic epitopes DNA vaccine in the eukaryotic expression system.The DNA sequences encoding metalloproteinases multi-antigenic epitopes were artificially synthesized.Then the sequences,with or without signal peptide linking,were linked to pIRESneo expression vector pIRESneo to construct two plamids for immuning mice.The result was detected by ELISA and the method of venom hemorrhagic activity neutralization.Both of two recombinant plasmids could induce antibody(1∶100) and relieve metalloproteinases-induced haemorrhage.Thus we can conclude that signal peptide dose not influence the metalloproteinases DNA vaccine expresession in mice.
