硫化氢通过SIRT1抗内皮细胞衰老 被引量:1

摘要 目的探讨外源性硫化氢在人脐静脉内皮细胞(HUVEC)衰老中的作用及分子机制。方法用25μmol/L H2O2诱导HUVEC衰老,通过检测β-半乳糖苷酶计算细胞衰老率。检测不同浓度(15、30、60及120μmol/L)的硫氢化钠作用下内皮细胞衰老标志物β-半乳糖苷酶的表达,Western blot分析沉默信息调节因子1(SIRT1)蛋白的表达。结果 HUVEC经25μmol/L H2O2处理1 h后,β-半乳糖苷酶阳性细胞率为12.2%±1.30%;经60μmol/L硫氢化钠干预48 h后,β-半乳糖苷酶阳性细胞率显著降低,为4.6%±1.14%,两组相比差异显著(P<0.05);与对照组比较,60μmol/L硫氢化钠干预48 h后SIRT1蛋白表达显著上调(P<0.05),而用SIRT1抑制剂(NAM)可减弱硫氢化钠此作用(β-半乳糖苷酶阳性细胞率为12.0%±1.58%)。结论硫化氢能通过上调SIRT1表达抵抗H2O2诱导HUVEC衰老。
