摘要
目的观察ZEA对离体培养HeLa细胞的生长抑制作用和对超微结构影响,探讨其毒性作用机制。方法用四甲基偶氮唑蓝(MTT)检测不同剂量ZEA暴露24 h后,对HeLa细胞生长抑制作用。在透射电镜下观察ZEA所致细胞超微变化情况。结果 Hela细胞生长抑制率组间比较,差异有统计学意义(F=1 099.093,P<0.05);任何两组间细胞生长抑制率比较,差异有统计学意义(P<0.05);细胞生长抑制率随着染毒剂量的增加而增加。电镜下可见低剂量组细胞胞质内线粒体空泡变性和髓样变;高剂量组坏死细胞数量明显增多,细胞核固缩、畸变、崩解,线粒体嵴絮状变。结论 ZEA对Hela细胞有明显的细胞毒性作用并可导致细胞超微结构损伤。
Objective To observe the effect of zearalenone(ZEA) on the growth inhibition and ultrastructure of HeLa cells,and to explore the mechanism of the cytotoxicity.Methods Methyl thiazolyl tetrazolium(MTT) was used to assay the growth inhibition of different doses of ZEA on HeLa cells after 48 h exposure and transmission electron microscope was used to observe the ultrastructure of HeLa cells.Results The inhibition rate of cell growth was significantly different between groups(F=1099.093,P0.05),were all significantly different between every two groups(all P0.05).The growth inhibition rate increased as the ZEA dose increased.Vacuolar degeneration and pulpiform change of mitochondria were seen in the cytoplasm in lower dose group under transmission electron microscope.The number of necrotic cells was increased in higher dose group,and nuclear pyknosis,aberration,nucleonic fragments and flocculent degeneration of mitochondria appeared.Conclusion The cytotoxic effect of ZEA on HeLa cells and ultrastructural damage was observed.
