摘要
目的探讨高血糖合并形觉剥夺性近视/弱视小鼠模型的制备方法。方法选取1周龄的BALB/cJ小鼠30只,通过查随机数字表分为3组,每组10只:正常对照组、诱发高血糖组、诱发高血糖联合形觉剥夺性近视/弱视组。右眼睑缝和建立形觉剥夺性近视/弱视,腹腔多次小剂量注射链脲菌素(STZ)诱发高血糖。结果成功建立形觉剥夺性近视/弱视与高血糖小鼠模型;与对照组相比,诱发高血糖小鼠的血糖[高血糖组(20.66±3.51)mmol/L,高血糖合并形觉剥夺性近视/弱视组(21.05±5.91)mmol/L]、体质量均显著改变[对照组(26.90±4.38)g,高血糖组(18.02±3.90)g,高血糖合并形觉剥夺性近视/弱视组(15.87±5.61)g;P<0.01]。缝合眼睑小鼠右眼眼轴长度、屈光度[(3.348±0.018)mm,(9.05±1.42)D]与对照组[(3.310±0.041)mm,(11.80±2.24)D]相比,差异有统计学意义(P<0.01)。结论缝合眼睑联合STZ小剂量多次腹腔注射是一种可靠的制备高血糖合并形觉剥夺性近视/弱视小鼠模型的方法。
Objective To establish a new animal model of hyperglycemia associated with form-deprivation myopia/amblyopia in order to provide animal experiment data for researching the relationship between early stage diabetic retinopathy and refraction error.Methods A total of 30 one-week-old BALB/cJ mice were randomly divided into 3 groups(n=10 for each group),control group(group Ⅰ),hyperglycemia group [group Ⅱ,repeatedly injected of 50 mg/kg streptozotoein(STZ) intraperitoneally from 30 d after birth to induce hyperglycemia],and hyperglycemia and form-deprivation myopia/amblyopia group(group Ⅲ,with their right eyes were sutured at 10 d after birth,and then injected with 50 mg/kg STZ in 30 d after birth).Their blood glucose level were measured from 1 d after STZ injection,once a week.Their dioptres were examined,and body weight and ocular axial length were measured in 82 to 87 d.Results Group Ⅱ and group Ⅲ had much higher blood glucose(20.66±3.51 and 21.05±5.91 mmol/L) and significantly lower body weight(18.02±3.90 and 15.87±5.61 g) when compared with those of control group(8.77±4.38 mmol/L and 26.90±4.38 g respectively,P0.01).Compared with group Ⅰ(11.80±2.24 D,3.310±0.041 mm) and group Ⅱ(12.25±1.77 D and 3.315±0.031 mm),the diopter was significantly decreased and the ocular axial length was elongated obviously in the right eyes of group Ⅲ(3.348±0.018 mm,9.05±1.42 D,P0.01).Conclusion Hyperglycemia associated with form-deprivation myopia can be induced in BALB/cJ mice successfully through STZ injection and eye lid suturing when the mice are within 10-day-old.
