下肢动脉硬化闭塞症模型的探讨 被引量:8

摘要 目的:探索动脉硬化闭塞症(ASO) 家兔模型的制作方法,为ASO 中西医结合临床和实验研究提供动物实验对象。方法:选择20 只新西兰雄性家兔,实验组和对照组各10 只。利用4F气囊导管损伤髂股动脉内膜,配合高脂、高胆固醇饲养和免疫损伤等因素诱导髂股动脉ASO 家兔模型,制作周期20 周。模型制作前后行股动脉造影并于实验结束后处死动物并行病理学检查,于模型制作过程中动态观察血脂、血胆固醇、内皮素(ET) 和一氧化氮(NO) 水平。结果:模型制作过程中,血脂、血胆固醇和血浆ET逐渐升高,而血浆NO逐渐降低。模型制作后股动脉造影显示髂股动脉高度狭窄,远端血管显影欠佳。病理切片显示典型的ASO变化,与人类ASO病理变化相似。结论:本实验方法可成功地诱导ASO模型,拟人性、重复性较好,制作周期较短,比较经济。 To establish the atherosclerosis obliterans(ASO)model in rabbits for the better basis and clinic study.Methods:20 Newzealand rabbits,male,2500~3000gram,were divided into two groups evenly at random.Group A was for experiments and group B for the control.The group A was raised by the fodder riched in glycerid and cholesterol for 20 weeks with the mechanic injuries and immune injuries,while the group B was raised by normal fodder for 20 weeks.All rabbits were examined by arteriography before and after the model making processes,and all rabbits were killed at the end of the experiment.All ilofemoral arteries were examined in pathologically.The total serum glyceride and cholesterol,plasma endothelins(ET)and nitric oxide(NO)were successive determined during the process.Results:Total serum glyceride and cholesterol,plasma ET were increased gradually,while the level of plasma NO was decreased during process.The iliofemoral artery stricture and ASO pathological signs which analoged to human ASO were found in arteriography and pathological examination.Conclusion:This relative economic and short period experiment method may induce successfully ASO rabbit models which have good personification and reiterration.
