摘要
[目的]观察NUMBL在大鼠脊髓损伤过程中的表达情况,探讨其在脊髓损伤和修复过程中的生物学功能。[方法]制作脊髓损伤模型,利用western blot、免疫组织化学、免疫荧光等技术观察NUMBL在损伤脊髓中的表达变化、组织分布与细胞定位,在此基础上探讨NUMBL在脊髓损伤后的病理生理意义。[结果]western blot结果显示NUMBL经历了在损伤后8 h和1 d的显著增加后开始下降,直到损伤后2周仍没有明显上调的变化趋势。免疫组化结果显示在脊髓损伤后1 d,NUMBL的表达在临近损伤中心的脊髓中显著增加,而在脊髓损伤后1周,NUMBL的表达较1d时明显降低。免疫荧光结果显示NUMBL与神经元和少突胶质细胞存在共定位,而与星形胶质细胞无明显共定位。[结论]脊髓损伤后脊髓中NUMBL的表达上调参与脊髓损伤的病理过程。
[Objective]To observe the expression patterns of NUMBL in rat models of spinal cord injury,and analysis the association with neuronal survival or apoptosis and glial proliferation,as well as to investigate their potential biological roles in nerve injury and regeneration.[Method]Sprague-Dawley(SD) rats were subjected to spinal cord injury.In the purpose of exploring the pathological and physiological significance of NUMBL,the expression patterns and distribution for NUMBL were observed by western blot,immunohistochemistry and immunofluorescence.[Result]Western blot analysis showed NUMBL protein level was significant up-regulated at 8 hours and 1 day,then reduced and had no up-regulation to 2 weeks.Immunohistochemistry analysis showed that the immunochemical staining was significantly increased at 1 day and then reduced at 1 week.Immunoflurescence analysis showed NUMBL were expressed in neurons and oligodendrocytes but not in astrocytes.[Conclusion]The up-regulation of NUMBL in the injured spinal cord may be associated with apoptosis process of neurons and oligodendrocytes.
