摘要
目的 观察体外培养的正常猪视网膜色素上皮 (RPE)细胞中游离钙。方法 培养猪RPE细胞后 ,用激光扫描共聚焦显微镜 (LSCM)观察细胞中游离钙的分布特点 ,并通过校正求得游离钙的绝对值。结果 猪RPE细胞中游离钙的分布特点是细胞核较细胞浆中浓度高 ,通过校正求得细胞中游离钙平均值为 ( 2 2 0 .3 7± 2 7.3 5 )nmol/L。结论 用LSCM能直接观察培养的RPE细胞内游离钙的分布 。
Objective To determine the free calcium in normal cultured porcine retinal pigment epithelial(RPE)cells. Methods The concentration of free calcium in cultured RPE cells was determined by laser scanning confocal microscopy(LSCM).Measurement of concentration of free calcium in RPE cells was determined by calibration. Results The free calcium in RPE cell nuclei was higher than that in their cytoplasm.The concentrations of free calcium in RPE cells were 220±27.35*!nmol/L. Conclusion LSCM can be used to observe the characteristics of free calcium in cultured RPE cells directly and obtain the concentration value of free calcium.
