摘要
以蚕豆(Vicia faba)根尖和玉米(Zaa mays)雄穗为材料,对染色体临时片制片方法进行了改良。结果表明,配置苏木精染色液时,加入微量的强氧化剂碘酸钠(NaIO3)可以使苏木精迅速氧化,达到即配即用的目的;在压片过程中,直接放盖玻片再用手压碎根尖、用镊子先将根尖压碎再放盖玻片,得到的制片效果好,且细胞分散均匀。
With the root tips of broad bean(Vicia faba,2n=12)and corn tassel as examination material,an improved method was used for observation of chromosome.The result indicated that during the process of Hematoxylin staining solution preparing,adding small amounts of NaIO3 can make rapid oxidation of Hematoxylin,which can be used to stain instantly now;In the course of chromosome squashing,the improved method in which materials were crushed by forceps firstly and then covered by slip is better than conventional method.
