摘要
目的 :比较益肾壮骨、健脾利湿和养血柔肝中药对原发 OA黑鼠关节软骨内几种细胞因子基因转录水平的影响。方法 :以运动负荷造成 C5 7黑鼠 OA模型 ,以 RT- PCR方法测定各组动物关节软骨内 IL- 1β、IL- 6、i NOS m RNA基因转录水平。结果 :C5 7黑鼠关节软骨 IL- 1β、i NOS m RNA表达随增龄逐渐增强 ,OA中期最明显 ,晚期下降。在 OA早、中期 ,益肾方可轻度下调关节软骨 IL- 1β、i NOS的基因表达 ,对 IL- 6有轻度上调作用 ;健脾方和柔肝方可明显下调 IL- 1β、i NOS的基因表达 ,与芬必得比较 ,有显著性差异 ,对 IL- 6无明显影响。结论 :不同治则方药对关节软骨细胞因子的基因表达影响不一 ;健脾方和柔肝方可明显下调鼠关节软骨 IL - 1、i NOSm RNA表达。
Objective:To compare the influences of the Kidney tonifying and bone strengthening herbs (KTH),the Spleen reinforcing and Dampness eliminating herbs (SRH) and the Liver softening and blood nourishing herbs (LSH) on several cytokines.Methods:By using RT PCR method,gene transcription of IL 1β,IL 6 and iNOS mRNA in the articular cartilage of the models of C57 black mice with osteoarthritis (OA) by sports loading was measured.Results:The gene expression of IL Iβ,iNOS increased distinctly in the articular cartilage of C57 black mice with OA,especially in the middle stage of OA.KTH can slightly down regulate the gene expression of IL 1β in the articular cartilage,and up regulate that of IL 6.SRH and LSH can apparently down regulate the gene expression of IL 1β and iNOS,but no obvious effect on that of IL 6 was observed.Conclusion:Different Chinese herbs have their respective effects on the gene expression of cytokines in articular cartilage.SRH and LSH can apparently down regulate the gene expression of IL 1β and iNOS.
