摘要
目的 建立简便、灵敏、试剂稳定性好的分光光度法测定血清锌。方法 在表面活性剂 Triton X100 存在下,用2(5溴2吡啶偶氮)5二乙氨基酚(5 Br P A D A P) 作显色剂测定血清锌。结果 该方法显色络合物最大吸收波长为558 nm ,线性范围达46 μmol/ L,表观摩尔吸光数为129 ×105 L·mol- 1·cm - 1 。回收率为983 % ~1016 % ,批内变异系数( C V) 和批间变异系数分别为24 % 与34 % ,与原子吸收分光光度法比较具有良好的相关性, Y= 1018 X- 0266r= 0998 5 , P> 005 ;56 例健康人血清锌含量为1037 ~2469 μmol L(珋x ±2s) 。结论 该法具有快速、简便、灵敏可靠等优点,适合临床应用。
Objective To establish a simple ,sensitive and stable method of determination of zinc inserum by U V spectrophotometry . Methods For determination of serum zinc with 5 Br P A P A Pin the presence ofsurfactant Triton X 100 . Results The maximum absorption ofthe complex compound was at558 nm . Thelinearitywas up to 46 μmol· L- 1 . The molar absorption coefficient was 1 .29 ×105 Lmol- 1·cm - 1 . The recoveries rangedfrom 98 .3 % to 101 .6 % . The withinrun and between run C Vwas 2 .4 % and 3 .4 % . Comparingthis method ( Y)with A A S( X) ,we obtainedthe regression equation . Y= 1 .018 X- 0 .226 , r= 0 .988 , P> 0 .05 . Thereferenceinterval(珋x + 2s) was 10 .37 ~24 .69 μmol· L- 1( n = 56) . Conclusion This method is simple ,sensitive andsuitablefor clinicalapplication .
