摘要
目的:通过观察第3腰椎横突综合征大鼠中枢不同部位八肽胆囊收缩素(CCK-8)mRNA阳性细胞表达的变化,探讨针刀疗法治疗第3腰椎横突综合征的中枢镇痛机制。方法:选用SD雄性大鼠,随机分为正常组、模型组、电针组和针刀组,后3组采用埋置明胶海绵的方法建立第3腰椎横突综合征的动物模型,电针组和针刀组分别给予电针左侧"肾俞"、"腰阳关"和针刀松解局部治疗。实验第28d进行取材,应用原位杂交方法检测各组动物下丘脑CCK-8 mRNA阳性细胞的表达。结果:造模后,大鼠脊髓背角、下丘脑和缰核CCK-8 mRNA的阳性细胞表达较正常增多,阳性强度增高,有显著性差异(P<0.01),电针组和针刀组中枢各部位CCK-8 mRNA阳性细胞表达较模型组减少,阳性强度降低,有显著性差异(P<0.05,P<0.01)。结论:针刀疗法和电针可通过降低中枢CCK-8 mRNA的表达,达到中枢镇痛的目的。
Objective:To observe the mRNA expression of CCK-8 in spinal dorsal,hornhypothalamus,habenular nucleus,and study the central analgesic effect of acupotomy in rats with the third lumbar vertebrae transverse process syndrome.Methods:SD rats were randomly divided into normal group,model group,electroacupunture(EA) group and acupotomy group.The rats were induced to the model by inserting a piece of gelatin sponge into the posterior of the left 3rd lumbar vertebrae transverse process.EA group and acupotomy group are used EA therapy applied to the left 'Shen shu','Yao yang guan' and local minimal invasive lysis respectively.The mRNA expression of CCK-8 in spinal dorsal,hornhypothalamus,habenular nucleus were assayed by in situ hybridization method on day 28.Results:Compared with normal group,the number of CCK-8 mRNA positive cells was increased significantly in spinal dorsal,hornhypothalamus,habenular nucleus in model group,positive stain intensity was increased significantly also(P〈0.01).In EA group and acupotomy group,the number of CCK-8 mRNA positive cells was decreased significantly in hypothalamus than model group,positive stain intensity was decreased too(P〈0.05,P〈0.01).Conclusion:Acupotomy and EA therapy could attain central analgesic effect by decreasing the mRNA expression of CCK-8 in central nervous system.
