摘要
目的探讨食管鳞癌中RUNX3蛋白表达及其与临床病理参数的关系。方法采用蛋白质印迹法检测RUNX3蛋白在42例食管鳞癌组织及癌旁组织中表达的差异,并分析与临床病理参数的关系。结果蛋白质印迹显示47.6%(20/42)的食管鳞癌组织RUNX3蛋白表达明显下调或缺失,而在癌旁正常组织中表达正常,RUNX3蛋白在食管鳞癌组织与癌旁正常组织中的表达差异有统计学意义(P<0.001),RUNX3蛋白的表达下调或缺失与淋巴结转移、TNM分期显著相关(P值分别为0.047及0.026)),与性别、年龄、肿瘤大小、分化程度、饮酒史、家族史等之间无明显相关(P>0.05)。结论 RUNX3蛋白在食管鳞癌组织中存在明显的表达缺失或下调,并与食管鳞癌TNM分期及淋巴结转移密切相关,其表达下调与食管鳞癌的浸润、转移有关。
【Objective】To evaluate expression of the RUNX3 protein and potential clinical implications in 42 ESCC tissues and matched 42 adjacent normal tissues.【Methods】To detect expression of RUNX3 protein by western blotting in 42 primary ESCC tissues and corresponding normal tissues,the results were compared with the clinicopathological data.【Results】Loss or down regulation expression of RUNX3 protein expression was detected in 20(47.6%) of the 42 ESCC tissues by western blotting analysis,whereas RUNX3 protein was detectable in all matched 42 adjacent normal tissues.There was significant difference of loss or down regulation of the expression of RUNX3 protein between ESCC tissues and corresponding normal tissues(P 0.001).Loss or down regulation of RUNX3 protein expression was significantly correlated with lymph node metastasis and TNM stage(P =0.047 and 0.026 respectively).No relationship was observed between the expression of RUNX3 protein and sex,age,tumor size,tumor differentiation or drinking(P 0.05).【Conclusion】Loss or down regulation expression of RUNX3 protein is frequent in ESCC,which is closely associated with lymph node metastasis and TNM stage.
