摘要 目的探讨IL-10和HO-1在酒精性脂肪肝模型中的作用和意义。方法建立大鼠酒精性脂肪肝动物模型,检测大鼠血清IL-10以及TNF-α,肝匀浆中MDA、GSH以及SOD相关指标的变化;此外采用RT-PCR方法检测IL-10和HO-1 mRNA的表达。结果随着肝脏损伤程度的加深,TNF-α的水平明显增加而IL-10却逐渐降低;肝匀浆中GSH和SOD的活性明显下降,但MDA却呈上升趋势;IL-10和HO-1mRNA表达也随着肝脏损伤程度的加深而减弱。结论 IL-10与HO-1的表达随着肝损伤程度的加重而逐渐降低,且构成一个新的抗炎途径参与了酒精性肝病的过程,从而为今后的酒精性肝病的临床治疗和药物研发提供新的思路。 【objective】To investigate the expression and significance of IL-10 and HO-1 in the animal model of alcoholic liver disease.【Methods】We establish an animal model of alcoholic liver disease(ALD).Meanwhile,alanine aminotransferase(ALT),aspartate transaminase(AST)、interleukin(IL)-10 and tumor necrosis factor(TNF)-from serum and GSH,SOD and MDA from liver homogenate were detected.And IL-10 and HO-1 mRNA were detected by RT-PCR.【Results】The levels of TNF-from serum were increased and the levels of IL-10 from plasma were decreased compared with those of normal group and control group.Thelow levels of GSH and SOD activities and the high level of MDA concentration from the liver homogenate were in the alcohol group compared with those of normal group and control group.Meanwhile,with the sever-ity of the liver injury,the expression levels of IL-10 and HO-1 mRNA were decreased.【Conclusion】The expression levels of IL-10 and HO-1 were decreased during the severity of liver injury.And they,as a newanti-inflammatory pathway,may participate in the process of ALD and provide some new ideas for the future clinical treatment and the drug research of ALD.
