摘要
将两株抗旋毛虫单克隆抗体(McAb)杂交瘤细胞(D4,E2)所分泌的McAb,经纯化后电泳分析,其重链分子质量为55ku,轻链分子质量为29ku,符合IgG抗体的特点。两株McAb与旋毛虫阳性猪血清所针对的旋毛虫抗原决定簇相同,都不与猪的其它寄生虫发生交叉反应,与抗原的相对亲和力E2>D4,McAb亲和层析纯化抗原的分子质量为49ku,经免疫斑点染色证明为糖蛋白。
The monoclonal antibodies (McAbs) against
[WTBX]Trichinella spiralis secreted by two strains of hybridomas (D 4,E 2) were purified and
analysed with electrophoresis, which revealed that the molecular masses of the heavy chain
and the light chain were 55 ku and 29 ku respectively which was in accord with the trait of IgG.
The two McAbs and the Trichinella spiralis postive pig's serum aimed at the same antigenic
determinant but no cross reaction with another parasites in swine occurred; the relative affinity
with antigen of E 2 was stronger than that of D 4. The molecular mass of McAb affinity
chromatography purified antigen was 49 ku. It was proved that the purified antigen was a
glucoprotein by means of an immunity dot staining test.
