缬沙坦和血管紧张素Ⅱ对人体内皮细胞表达纤溶因子的影响 被引量:1

摘要 目的:探讨血管紧张素Ⅱ(Ang Ⅱ)和AngⅡ 1型受体拮抗剂缬沙坦对人体内皮细胞纤溶酶原激活因子(纤溶因子)表达的影响.方法:选用人脐静脉内皮细胞系(HUVECs)作为研究对象.采用不同浓度AngⅡ(10-5~10-9mol/L)与HUVECs共同培养,作为AngⅡ组,同期设立对照组和缬沙坦组.运用细胞酶联免疫法和发色底物法检测三组细胞培养上清液中组织型纤溶酶原激活剂(tPA)和I型纤溶酶原激活物抑制剂(PAI-1)的活性与抗原浓度.结果:AngⅡ呈浓度依赖型刺激PAI-1的活性和含量水平,缬沙坦可降低AngⅡ对PAI-1分泌的作用.二者对tPA的分泌均无明显影响.结论:AngⅡ可促进HUVECs分泌PAI-1;缬沙坦可阻断Ang II的这种作用. Objecitve:Discusses the blood vessel tense elementⅡ(Ang Ⅱ ) And Ang Ⅱ1 acceptor antagonist compound patterned silk Sha Tan (filament dissolves the influence to the human body endothelial cell plasmin original activatio'n factor which factor) expresses. Methods:Selects human umbilical vein bast cell line (HUVECs) to take the object of study. Uses different density AngⅡ(10 - s 10 - 9 mol/L) raises together with H UVECs, takes AngⅡThe group, sets up the control group and patterned silk Sha Tanzu same time. And sends the color substrate law using the cell enzyme association immunity law to examine in three group of cell raise supernate organization plasmin original activating agent (tPA) and I plasmin stirs up the living creature inhibitor (PAI-1) activeness and the antigen density originally. Results: AngⅡAssumes the density dependent form to stimulate PAI-1 the activeness and the content level, has blurred vision Sha Tanke to reduce AngⅡTo PAI-1 secretion function. The two to tPA secretion not obvious influence. Conclusion: AngⅡMay promote HU- VECs to secrete PAI- 1 ; Patterned silk Sha Tanke blocks the Ang Ⅱthis kind of function.
