摘要
目的探讨骨髓间充质干细胞(MSC)对大鼠急性肺损伤的作用及机制。方法雄性Wister大鼠4只,用于骨髓间充质干细胞采集;分离培养大鼠骨髓间充质干细胞,传致第Ⅳ代备用。雌性Wister大鼠64只,随机分为对照组(C组)、内毒素肺损伤组(LPS组)、骨髓间充质干细胞治疗大剂量组(MSCH组)和小剂量组(MSCL组)。C组和肺损伤模型经气管内分别注入生理盐水和内毒素,1小时后,C组和LPS组大鼠经尾静脉注入生理盐水,MSC组(MSCH组和MSCL组)大鼠经尾静脉注入骨髓间充质干细胞混悬液2 mL(1×106和1×105两个剂量)。分别在24 h和72 h留取标本,检测肺组织形态学、肺组织湿干重比、支气管肺泡灌洗液(BALF)白细胞总数、蛋白含量、肿瘤坏死因子-α(TNF-α)和白细胞介素-8(IL-8)水平。结果 LPS组和C组比较,肺组织病理评分、湿干重比、BALF中白细胞总数、蛋白含量、TNF-α及IL-8水平显著升高;MSC两组较LPS组各炎症指标均有明显改善,且MSCH组较MSCL组效果显著。结论 MSC移植可显著减轻内毒素诱导的急性肺损伤,且这种作用强度是剂量相关的。
Objective To explore the effect and its mechanism of bone marrow mesenchymal stem cells(MSC) on rats with acute lung injury.Methods Four male Wister rats were used for collecting MSC.Female Wister rats were randomly divided into control group(C group),endotoxin lung injury group(LPS group),and MSC high-dose group(MSCH,1×106) and low-dose group(MSCL,1×105).2 mL MSC was infused intravenously and intratracheal administration of LPS one hour later.At 24 h and 72 h,samples were collected for assessment of histology,biochemical analysis and levels of TNF-α and IL-8 in bronchoalveolar lavage fluid(BALF).Results Compared with LSP group,there was a significant decrease in lung wet to dry ratio,lung injury score,WBC counts,protein levels,TNF-α and IL-8 in BALF in the MSC-treated rats at 24 h and 72 h,and the MSCH group was better than the MSCL group.Conclusion Treatment with MSC significantly reduces endotoxin-induced acute lung injury and the effect is related to its dose.
