摘要
目的探讨幼鼠肾间质纤维化模型(单侧输尿管结扎模型)中基质细胞衍生因子-1(SDF-1)不同时间点的表达趋势、相关性,以及用阿托伐他汀干预的效应。方法建立幼年大鼠肾间质纤维化模型。将90只幼鼠随机分为对照组、药物组和模型组,每组各30只。各组分别于实验第1、3、5、7、14天各时间点取肾组织进行相关指标测定。采用Masson的染色方法评价各个时间点肾小管受损的程度。免疫组织化学法检测大鼠肾组织中SDF-1和转化生长因子(TGF)-β1的蛋白表达趋势及相关性。结果对照组、模型组和用药组的SDF-1灰度值相比较,显示3组之间差异有统计学意义(F=9.04,P=0.001),且对照组与模型组和用药组差异有统计学意义(P<0.05),3组各个时间点的比较结果显示,模型组(F=70.86,P<0.01)和用药组(F=92.28,P<0.01)不同时间点的灰度值之间差异有统计学意义,模型组与用药组在各时间点间的灰度值比较结果除了第1天差异无统计学意义(P>0.05),其余各时间点之间差异有统计学意义(P<0.05)。TGF-β1的结果与SDF-1类似。结论在幼年大鼠肾间质纤维化过程中,肾间质内SDF-1表达上调对肾间质纤维化进程可能有抑制作用;而阿托伐他汀干预后有明显的干预效应。
Objective To explore the trends and relevance of stromal cell-derived factor-1(SDF-1) expression in young rats with renal interstitial fibrosis induced by unilateral ureteral obstruction(UUO) at different time spots,and the effects of atorvastatin intervention.Methods The renal interstitial fibrosis models were established with young UUO rats.Ninety rats were selected and randomized into control group,atorvastatin group and model group(n=30 each).The kidney tissues were obtained and examined for certain laboratory indexes in each group on days 1,3,5,7 and 14.The degree of tubulointerstitial injury at these time spots in the three groups was evaluated by Masson staining.The expression trends of SDF-1 and TGF-β1 proteins were detected by immunohistochemistry,and analyzed for the correlation between each other.Results There was a significant difference in gray values of SDF-1 across the control,model and atorvastatin groups(F=9.04,P=0.001),between the control group and model group,and between the control group and atorvastatin group(both P0.05).At all time spots,the gray values of SDF-1 were different in the model group(F=70.86,P0.01) or in the atorvastatin group(F=92.28,P0.01).At any given time spots,the gray values of SDF-1 were different between the model and atorvastatin groups(P0.05) except on day 1(P0.05).The outcome of TGF-β1 was similar to that of the SDF-1.Conclusion In the process of renal interstitial fibrosis,up-regulated expression of SDF-1 in renal interstitium may have an inhibitory effect on the progression of renal interstitial fibrosis.Atorvastatin may exert a significant intervention effect.
