摘要
为评价基因重组转化生长因子—β(TGF—β)对骨折愈合的促进作用,采用基因重组的方法提取转化生长因子—β,体外检测其活性后,用于兔股骨骨折模型。采用新西兰大白兔20只,同体双侧对照,将重组转化生长因子—β100μg与纤维蛋白凝胶制成复合物随机植入股骨骨折线两端的髓腔中,将等量的单纯纤维蛋白凝胶植入对侧肢体中,术后2、4、6周三阶段做X线平片观察,放射性核素γ骨显像进行骨折区放射性计数,组织学观察。结果显示:使用重组转化生长因子—β侧肢体在骨折区放射性计数、组织形态学上在各阶段都优于对照侧肢体。认为重组转化生长因子—β具有明显的促进骨折愈合的作用。
The purpose was to evaluate the effect of gene recombinant TGF - βon the induction of bone formation and stimulating fracture healing. In this experiment, TGF -βwas extracted by gene recombinant method.Twenty Newzealand white rabbits were used, and animal model of bilateral femur fracture were established with internal fixation of 4 hole plate and screws. In marrow cavity of both end of fracture line, 100μg TGF-β and fibrin was inserted in the treated side, the fibrin alone was inserted in opposite side as control. After the operation, the formations and healing of bones were determined by serial radiographic, histologic, SPECT analysis.Results showed that the radioactive gauges of fractures area's in treated side were higher than those in control side. Authors consider that the gene recombinant TGF-βenhanced the fracture healing obviously.
