ox-HDL_3对人脐静脉内皮细胞的损伤作用 被引量:2

摘要 目的:探讨氧化高密度脂蛋白3(ox-HDL3)对人脐静脉内皮细胞(HUVECs)的损伤作用。方法:体外培养HU-VECs与不同浓度(50~200mg/L)ox-HDL3共孵育,用普通光镜和荧光显微镜分别观察HUVECs形态及核形态的变化,用噻唑兰(MTT)比色法检测细胞存活状况。结果:ox-HDL3使内皮细胞数目减少,形态改变;Hoechst 33258染色后可见大量的细胞核浓缩呈高亮度蓝色荧光的凋亡细胞;MTT结果显示,细胞存活率下降,且呈剂量依赖性。结论:ox-HDL3可导致内皮细胞受损,从而促进动脉粥样硬化的发生发展。 Objective:To explore the effect of ox-HDL3 on human umbilical vein endothelial cell(HUVEC).Methods:HUVECs were incubated with ox-HDL3(50、100、200mg/L) for 24 hours.The change of cell shape and nuclear morphology were observed by light microscope and nuclear staining with Hoechst 33258.The cells survival percentage was detected by MTT assay.Results:Compared with the control group,the number of HUVECs cells incubated with ox-HDL3 decreased and the shape changed.Cells appeared typical characteristics of apoptosis,including apoptotic nuclear condensation.The survival rate of cells was reduced by MTT assay.Conclusion:ox-HDL3 can lead to HUVECs damage.
