摘要
目的探讨急性白血病(AL)病人X染色体连锁凋亡抑制蛋白(XIAP)的表达及临床意义。方法采用逆转录聚合酶链反应技术检测了46例不同类型、不同阶段AL病人白血病细胞中XIAP的表达情况,以11例健康人作为对照。结果 AL病人白血病细胞XIAP表达水平比对照组明显增高(t=13.96,P<0.01)。初治组、缓解组和复发组AL病人白血病细胞XIAP表达水平均明显高于对照组(F=204.27,q=8.88~32.69,P<0.01),其中缓解组明显低于初治组和复发组(q=12.44、26.02,P<0.01),复发组明显高于初治组(q=17.55,P<0.01)。结论 XIAP表达可能参与了AL的发生和发展过程,XIAP高表达的病人完全缓解率低,预后不良。XIAP高表达可能是导致白血病细胞对化疗药物不敏感的原因之一,XIAP有望成为白血病基因治疗的新靶点。
Objective To discuss the expression of X-linked inhibitor of apoptosis protein(XIAP) and its clinical significance in patients with acute leukemia(AL). Methods By using semi-quantiative reverse transcriptase polymerase chain reaction,the expression of XIAP in 46 AL patients of various types and at different stages was detected,and 11 healthy persons served as normal controls. Results The levels of XIAP expression in AL patients in three groups,i.e.initial-treatment group,remission group,and recurrence group,were higher than that in normal controls(F=204.27,q=8.88-32.69,P0.01),in which,XIAP expression of those in the remission group was lower than that in the initial-treatment group and recurrence group(q=12.44,26.02;P0.01);that in recurrence group was higher than that in initial-treatment group(q=17.55,P0.01).Conclusion The expression of XIAP is involved in the occurrence and development of AL.The patients who have over expression of XIAP will experience a low complete-remission rate and poor prognosis.Over expression of XIAP is likely to be one of the reasons for leukemic cells resistant to chemotherapeutics,which is hopefully to become a new target of gene therapy for leukemia.
