蛋白质相互作用技术在热带植物病毒研究中的应用 被引量:1

摘要 蛋白质-蛋白质相互作用(Protein-protein interaction)承载着生物体生命活动的各个过程,研究热带植物病毒中的蛋白质-蛋白质相互作用,不仅可以从分子水平揭示病毒蛋白质的功能,还可以揭示病毒在寄主细胞中的侵染机理,从而制定相关策略预防病毒的突然爆发或进行疾病治疗。而蛋白质相互作用研究方法已有50多年的历史,如早期的化学交联法;并且该方法的技术在不断地完善和更新,产生了一序列新的技术,包括酵母双杂交、GST pull-down,免疫共沉淀和串联亲和纯化等多种技术。由于本实验室在热带植物病毒研究中涉及了多种蛋白质相互作用技术,为此,本文总结热带植物病毒研究中常用的蛋白质相互作用技术。 Protein-protein interactions control the processes of life activities of organisms.Research of protein-protein interactions in tropical plant virus not only reveals the function of viral proteins in molecular level,but also reveals the mechanism of virus infected host cells.Finally,relevant strategies are made to prevent sudden outbreak of virus or to treat diseases.Methods of investigation of protein interactions such as chemical crosslinking former began more than 50 years ago.The methods keeps improving and updating and a series of new technologies,including yeast two-hybrid,GST pull-down,immunoprecipitation,tandem affinity purification and other technologies have been originated from them.This paper summarized some protein interaction technologies which are commonly used in tropical plant virus in our laboratory.
