甲醇营养型酵母Pichia pastoris在近十几年已被人们广泛用作外源基因表达的系统。表达的是可溶性蛋白,且胞外分泌。本研究系将外源基因(胰岛素原基因)连接到穿梭质粒PHIL-S1上,再通过同源重组到酵母染色体,筛选表达株。用1 升发酵罐在甲醇诱导下可获得0.
Methylotrophic yeasts Pichia pastoris is faster,easier,and less expensive to use than other eukaryotic expression system such as baculovirous or mammalian tissue culture,and generally gives higher expression levels.It can express heterologous soluble protein and secret out of cell.Clone foreign gene(pro insulin gene) into PHIL\|S1,and recombinate onto yeast chromosome.In yeast Pichia pastoris, under the induction by methanol,the expressed proinsulin was secreted into medium at the level of 300mg/L.\;
